Tetracyclines are a class of synthetic antibiotics with the ability to induce gene expression in cells in a process called transactivation and the development of cell lines and systems. We showed that tetracycline (TA) binds to an intracellular promoter element in the promoters of various genes that contain the TTR promoter. Tetracycline has a low affinity for the TTR promoter, and this binding is reduced when the promoter is deleted. A series of experiments investigated the binding of TA to the promoter and the induction of the expression of genes in transgenic mice. The results showed that the induction of the expression of genes in transgenic mice is inhibited when the promoter is deleted.
